Kingpin's clear interest in a political campaign hints at an adaptation of Marvel's Devil's Reign comic book storyline, which would possibly mark the highest point in Wilson Fisk's MCU journey.
In Devil's Reign, Wilson Fisk uses his influence and power as NYC mayor to outlaw all superheroes in the city. Fisk's decision causes all New York superheroes to join forces against him, including the Fantastic Four, Moon Knight, Elektra, Daredevil, Jessica Jones, and Luke Cage. While an MCU adaptation of Devil's Reign would inevitably look quite different in live-action, it could still incorporate some of the comics' key players like Luke Cage, who becomes Fisk's political opponent in the original storyline, but became a crime boss in the Luke Cage season 2 finale.

Kingpin already has the ideal justification to achieve it: heroes such as Daredevil, Spider-Man, the Punisher, and Luke Cage work outside the law and remain unregulated after the Sokovia Accords became a failure. Since Luke Cage was last seen becoming Harlem's top crime boss and taking over Harlem's Paradise, he's the perfect target for Kingpin to paint superhumans and vigilantes in a bad light to the public.

Which Street-Level Heroes Kingpin Seems Most Likely To Target Kingpin's political goals may be a form of revenge against Daredevil, Hawkeye, and Echo, who Wilson Fisk could persecute as soon as he can. However, Kingpin might wish to begin with easier targets. As a literal crime lord, Luke Cage provides Kingpin with the perfect example of superhumans using their abilities to break the law, despite the fact that Cage took over Harlem's Paradise precisely in order to stop actual criminals before they gain traction. Similarly, the Punisher only murders wrongdoers, but his crimes are easy to portray as the product of a mass murderer's irrational fixation on bloodshed.

After some success persecuting MCU street-level heroes, and with the help of journalists and conspiracy theorists like J. Jonah Jameson, Kingpin can easily manipulate the public's perception of heroes as powerful as Spider-Man. Despite his power, Spider-Man might be more vulnerable to grounded threats like Kingpin due to his recent lack of resources. Likewise, Jessica Jones, Iron Fist, Colleen Wing, Misty Knight, and Claire Temple could also be targeted and slandered by Kingpin, in part due to their illegal activities, and in part due to their connections to Daredevil.
What represents a dangerous superhuman or vigilante for Kingpin is unclear, however. Doctor Strange and Wong are known Avengers based in New York, but stopping a small crime or simply using their magic in public could place them beside Daredevil and Luke Cage. Other Avengers like Ant-Man, Hulk, and Bucky Barnes could also be detained temporarily if they show up in New York while Wilson Fisk is in power. Perhaps the only heroes who could avoid legal issues in New York could be James Rhodes a.k.a. War Machine and Sam Wilson a.k.a. Captain America, who represent the US Air Force and the government, respectively.
